Market Highlights: Neuropharmaceutical Approvals Down in 2009

In 2009, five of the 27 new drugs and biologics approved by the FDA were neuropharmaceuticals: Vanda/Novartis’ Fanapt for schizophrenia, Lundbeck's (Ovation) Sabril for infantile spasms and as an adjunctive for refractory partial seizures, Cypress/Forest’s Savella for fibromyalgia, Merck's (Schering-Plough/Organon) Saphris for bipolar and schizophrenia, and Novartis’ branded version of betaseron for multiple sclerosis. This compares to seven novel neuropharmaceutical approvals out of 24 in 2008 (Pristiq, Relistor, Entereg, Xenazine, Vimpat, Banzel and tapentadol) and only two of 17 in 2007. Acorda (ACOR) was the first to score an approval in 2010 for Ampyra (dalfampridine) to improve walking in multiple sclerosis (see story, page 6)...

Alzheimer's and Other Dementias: Treatments in Development
Dementia is a chronic or persistent deterioration of intellectual functioning, memory, executive and cognitive skills that is severe enough to interfere with daily life. Longer life expectancy has given rise to an increase in the number of dementia cases worldwide. Although most people, including many physicians equate dementia with AD, a disease that accounts for up to half of all dementias, there are in reality four common dementia syndromes with distinct pathological features. Unfortunately, the clinical distinctions between the common syndromes are subtle and vague and differential diagnosis is difficult. The four syndromes that account for almost all irreversible cases of dementia are Alzheimer's disease (AD), vascular dementia (VaD), Lewy body dementia (DLB), and frontotemporal dementia (FTD). The first two account for over 80% of irreversible cases. AD is the main target of current treatments in development. This article gives an overview of the different types of dementias and promising treatments in development ...

EnVivo Pharmaceuticals: Attacking Alzheimer's on Mulitple Fronts
EnVivo Pharmaceuticals of Watertown, MA was founded in the San Francisco Bay Area in 2001. It is dedicated to discovering and developing drugs for the CNS with an emphasis on neurodegenerative conditions and schizophrenia. The company has raised approximately $125 million from its sole investor Fidelity Biosciences over four rounds, the first of which closed in 2002. EnVivo slowly but surely evolved from an entity based on a discovery platform to a business with compounds in the clinic for Alzheimer's. While it is unlikely all of EnVivo’s trials will succeed given the track record of novel compounds for Alzheimer's treatment, it is clear that a multi-pronged, diverse pipeline strategy like EnVivo’s has the most chance of success...
